New Avenues Open Up for Mesothelioma Targeted Therapy

Newswise — July 25, 2011 (Philadelphia, PA) --Researchers from the lab of Antonio Giordano, M.D., Ph.D., the Founder and Director of the Sbarro Institute for Cancer Research and Molecular Medicine, have identified new potential anti-tumor agents that might be effective in treating mesothelioma, one of the deadliest cancer tumors.

Scientists tested new pyrazolo [3,4-d ]pyrimidine derivative inhibitors of the SRC kinase, a well-established molecular target in cancer therapy. They found that these SRC inhibitors effectively induced cell death, through apoptosis, in mesothelioma cell lines without affecting the normal mesothelial cells, thus supporting a possible use of these agents as a safe treatment for mesothelioma.
Their findings appear in the journal Oncogene.

Interestingly, the researchers also found that the SRC inhibitors induced cell death was accompanied by an increase in the nuclear stability of the cyclin-dependent kinase inhibitor p27. This is “particularly intriguing considering that the loss of nuclear p27 expression is a well established adverse prognostic factor in mesothelioma and p27 nuclear localization is crucial for its tumor suppressive function.” said the leading author of the study, Paola Indovina, Ph.D. of the University of Siena and assistant Professor at the Sbarro Institute.

“We think that these data represent a timely contribution and suggest that p27 status should be carefully analyzed when evaluating the use of kinase inhibitors affecting SRC in clinical trials for patients with mesothelioma,” said the corresponding author of the study Francesca Pentimalli, Ph.D. of the National Cancer Institute- “Pascale Foundation” - CROM- Cancer Research Center of Mercogliano in Avellino, Italy and assistant Professor at the Sbarro Institute.

“The findings support SRC as a critical therapeutic target in mesothelioma and reveal a new mechanism, dependent on p27 nuclear stabilization, by which SRC inhibition can induce apoptosis in mesothelioma cell lines, providing a new rationale for the use of SRC inhibitors in mesothelioma therapy,” says Prof. Antonio Giordano, another lead author. “It also shows that in this context, p27 is required to induce apoptosis in mesothelioma cell lines and, although this mechanism still needs to be precisely dissected, it adds further evidence supporting an active role for p27 in mediating apoptosis.”

The study was done in collaboration with Maurizio Botta full Professor of Medicinal Chemistry and Dean of the Faculty of Pharmacy, University of Siena, also adjunct Professor at the Sbarro Institute, who developed these new SRC inhibitors along with Prof. Silvia Schenone of the University of Genoa, Genoa, Italy.

Few would believe the effects of asbestos would span decades, affecting thousands and creating million dollar lawsuits. Litigators have documented asbestos lawsuits into waves.
Each wave is unique in the type of exposure, but unfortunately the results remain fatal.

The first wave of asbestos exposure began in the early 1900’s as manufacturing employees worked with heavy volumes of asbestos.
They worked directly with raw asbestos to create asbestos products for other industries.

It wasn’t until the second wave beginning in 1960, that manufacturers were forced to acknowledge the harmful substance.
Plaintiffs were primarily asbestos insulators for the construction and repair of military ships used in World War II.

Workers with direct contact suffered almost immediate health issues.
However, not only did they suffer but they passed the threat to family members.
Workers who came home wearing asbestos-tainted clothing would innocently hug or touch their family members, including their children.
This phenomenon is called take-home secondhand exposure.
The low exposure in these situations continues to increase the number of the third wave asbestos cases.

In the fourth wave of asbestos lawsuits, plaintiffs include construction and maintenance employees who worked with asbestos products.
Much of their exposure came from different job sites or in buildings where asbestos materials were used. This wave also includes those living or working in areas where asbestos products were installed.

These waves tell us that asbestos is not a relic from the past; it still afflicts many people, young and old, and with deadly consequences.
Attitudes towards asbestos related diseases must change if we are to ever see an end to it. Fortunately passionate nonprofit foundations, such as the Mesothelioma Applied Research Foundation and the Center for Asbestos Related Diseases, are dedicated to funding research to cure those suffering from asbestosis and mesothelioma.

San Francisco mesothelioma lawyers at The Asbestos Legal Center led a group of citizens affected by mesothelioma to meet with members of Congress on June 23.
The group, which was composed of Asbestos Legal Center’s attorney Raymond D. Mueller, a United States Navy veteran, a mesothelioma patient and his wife and several families who have lost loved ones to the disease, met with Senators Dianne Feinstein and Barbara Boxer on Capitol Hill.
The group asked the Senators to support legislation authorizing more money for mesothelioma research to help find a cure for the deadly disease.

Mr. Mueller asked the Senators from his home state to create an annual dedicated funding stream of five million dollars for mesothelioma.
Together with Asbestos Legal Center and its principal, Michael J. Mandelbrot, they have been representing victims of mesothelioma, lung cancer and asbestosis for more than 17 years.

Mesothelioma is a rare, incurable cancer caused by exposure to asbestos.
Many of those diagnosed with mesothelioma were exposed to asbestos serving in the United States Navy, or were exposed to asbestos performing construction and trade work.
It is estimated that 40 percent of the American labor force, or an estimated 27 million people, were exposed to asbestos between 1940 and 1980 and are considered at risk for developing mesothelioma symptoms. Researchers indicate that approximately 600 to 1,000 Armed Forces veterans die every year from mesothelioma and most of these individuals served in the United States Navy.

“We lose 600 to 1,000 veterans every year as a result of mesothelioma caused by exposure to asbestos in the military.
If it were a human enemy killing so many of our nation’s heroes, we would spend far more than five million dollars to try and save them.
Veterans from Sacramento and Stockton, to Los Angeles and San Diego, are dying, or have died, from asbestos-caused diseases at an alarming rate.
How many veterans will our government allow to die in the future before we start funding research at an adequate level to confront this disease?”, asked Mr. Mueller.

New York City mesothelioma lawyer Joseph W. Belluck, who is experienced in asbestos exposure issues, said he was disturbed to learn that a Brooklyn hospital has been cited for negligent failure to protect workers from exposure to the well-known carcinogen during demolition and renovation work.

The U.S. Department of Labor’s Occupational Safety and Health Administration has proposed fines of up to $48,000 against Interfaith Medical Center of Brooklyn for 14 violations of workplace health and safety standards during work to remove asbestos at the facility.

Among other infractions, the hospital failed to train staff, warn contractors of the presence of asbestos, properly label asbestos-containing materials and dispose of asbestos properly, according to an OSHA news release, which detailed the citations and the proposed penalties against Interfaith Medical Center.

“The connection of asbestos and asbestos removal to cancer, and the safety measures required are longstanding and well known,” said Belluck of the New York City personal injury law firm, Belluck & Fox, LLP. “It’s shocking that Interfaith Medical Center would ignore them.”

Last week, the Department of Defense (DoD) released the program announcement for the Peer Reviewed Cancer Research Program (PRCRP) for the 2011 grant cycle.
This will be only the fourth cycle to include mesothelioma as an eligible disease. Mesothelioma was included in the Peer Reviewed Medical Research Program in 2008, 2009 and 2010.
It was moved to the Peer Reviewed Cancer Research Program in 2011.

Four DoD grants were awarded at the end of 2009, and an additional 2.6 million was awarded in 2010 for a total of approximately 6.8 million awarded to mesothelioma research.

While this is momentous progress for mesothelioma research, the Meso Foundation continues to advocate for increased federal funding for mesothelioma and has, in fact, already testified in Congress for inclusion of mesothelioma in fiscal year 2012.

Ms. Hesdorffer explained the need for sustained and increased funding for mesothelioma through the Congressionally Directed Medical Research Program, and stressed the strong link between mesothelioma and military service.
She asked that a dedicated funding stream of $5 million be appropriated for mesothelioma through the Congressionally Directed Medical Research Programs, because mesothelioma disproportionally affects American servicemen and women and their families.

“Almost 3,000 Americans die each year of mesothelioma, and one study found that one third of patients were exposed on U.S. Navy ships or shipyards. That is 1,000 U.S. veterans and shipyard workers per year, lost through service to country, just as if they had been on a battlefield,” said Ms. Hesdorffer.

The Defense Subcommittee will be crafting the Defense Appropriations bill for FY 2012 in the next month, hopefully before the August Congressional recess.

In a significant victory for asbestos victims, including Timothy and Caroline Vest, Judge Eduardo C. Robreno, Jr. of the Asbestos Multi-District Litigation (MDL) Court (United States District Court, Eastern District of Pennsylvania, MDL 1935, Case No. 2:11-CV-63520) in Philadelphia granted plaintiffs’ motion to remand on May 26, 2011.
He ordered that the Vests’ asbestos personal injury case (Alameda County Superior Court, Case No. RG09489518) be immediately transferred back to Alameda County Superior Court.

Sign of Change for Asbestos Law
The Asbestos MDL Court was established in 1991 as the home for all asbestos lawsuits in U.S. federal courts. It currently consists of 60,000 cases and 3.5 million claims.
Due to this extremely high volume of cases pending before this court, several years can go by before a case is examined and sent back to state court or the local federal district court for trial.

Defendant McDonnell Douglas Delayed Case
On January 6, 2011, only 39 days before trial was to start in Alameda County Superior Court, McDonnell Douglas Corporation, the defendant, moved the Vests’ case to federal court. McDonnell Douglas claimed that while it knew about Timothy’s asbestos exposure claim relating to civilian planes the company manufactured, it did not know until December 2010 that he was exposed to asbestos from McDonnell Douglas’ military aircraft. The company claimed that the alleged source of asbestos fibers was enough to turn this into a federal case.
When McDonnell Douglas moved the case to federal court, it delayed the case for more than four months and caused the Vest family significant hardship, says Kazan. Because of the judge's quick action, his clients were not made to wait years, as some others have done.

The Judge’s Decision
Judge Robreno found that the removal (move to another court) was far too late. He held that McDonnell Douglas’ basis for removal was established on August 19, 2010---140 days before the removal. Timothy Vest’s father had testified on that date about work done on McDonnell Douglas military aircraft in the same hangar where Timothy Vest was exposed to asbestos.

Federal law only allows 30 days to remove once a defendant learns of a reason to do so. By wating 140 days, McDonnell Douglas sacrificed any basis for moving the case to a federal court and was ordered back to the Alameda court. Plaintiffs praised Judge Robreno’s decision. Said Kazan, “His very thorough scrutiny of the evidence before him led to the correct conclusion that McDonnell Douglas’ reason for removal was meritless.”

Mesothelioma, a cancer of the lining of the lungs, is closely linked to breathing asbestos, Belluck said.

According to the new study in Respirology, the journal of the Asian Pacific Society of Respirology, the increase in asbestos use in Asian countries since 1970 is likely to trigger a surge in asbestos-related diseases, including mesothelioma and lung cancer, in the next 20 years.

Asia’s share of worldwide asbestos use has steadily increased from 14 percent in the decades before 1970s to 64 percent from 2001 to 2007, the study said. Asbestos is widely used in building materials, roofing, cement and power plants in Asia. Yet many Asian countries that import asbestos have weak or non-existent workplace safety laws to protect workers from inhaling the toxic asbestos dust.

The portion of mesothelioma deaths recorded in Asian countries is far less than the percentage of asbestos used in Asia, suggesting mesothelioma deaths will rise significantly in the next 20 years. Workers exposed to asbestos typically take 20 to 50 years to show symptoms of malignant mesothelioma after inhaling asbestos fibers.

“Mesothelioma is a tragedy in slow motion, but the consequences are very real to the affected families,” Belluck said. “We believe a worldwide ban on asbestos, as called for by the World Health Organization, is the only sure way to stop the suffering and deaths caused by mesothelioma and asbestos-related disease.”

A new study that will look at how mesothelioma affects patients’ emotional and physical well-being could provide a more complete picture of the disease’s toll on victims and families, New York mesothelioma attorney Joseph W. Belluck said today.

New York’s Memorial Sloan-Kettering Cancer Center announced last week that it is conducting an investigation into how mesothelioma patients cope with the deadly disease in hopes of devising ways to reduce their psychological and physical strains.

“During the years I’ve spent fighting for the legal rights of people who contracted mesothelioma from asbestos exposure, I’ve seen what a crushing effect the disease can have both on the victims and their loved ones,” said Belluck, a New York asbestos exposure attorney and partner of Belluck & Fox, LLP.

Mesothelioma is an aggressive asbestos-related cancer that affects the lining of the lungs. The disease doesn’t respond to many standard cancer treatments, and victims usually face a dire prognosis — often living only 4-18 months after diagnosis.

Many victims of mesothelioma contracted the disease because they were exposed to workplace asbestos, according to Belluck.

“As you might expect, patients often react to the grim news of a mesothelioma diagnosis by becoming depressed and overwhelmed at the weight of it all,” the New York lawyer said. “The same is true for their loved ones and families.”

The Memorial Sloan-Kettering Cancer Center study is entitled “Mesothelioma from a Patient Perspective: A Survey of Psychosocial Needs and Exploration of Online Support for Patients.” One of its aims is to evaluate the possibility of developing ways of delivering emotional support to mesothelioma patients over the Internet.
Belluck, the New York mesothelioma lawyer, said he welcomed any developments that could improve the quality of life for the roughly 3,000 Americans who are diagnosed with mesothelioma every year.

Asbestos causes cancer. That was again the official word from a congressionally mandated study of cancer-causing substances released this month, New York mesothelioma attorney Joseph W. Belluck said today.

The 12th edition of the U.S. Department of Health and Human Services’ Report on Carcinogens found “asbestos and all commercial forms of asbestos are known to be human carcinogens based on sufficient evidence of carcinogenicity from studies in humans.”

“Asbestos has been listed as cancer-causing in every Report on Carcinogens since the first report was released in 1980,” said Belluck, the New York asbestos lawsuits attorney. “Although it’s been clear for more than 30 years that asbestos causes mesothelioma and other cancers, many victims are just now finding out that they have asbestos-related cancer because the disease can hide for decades.”

The 12th Report on Carcinogens said that studies have found that asbestos causes both pleural (lung) and peritoneal (abdomen) mesothelioma, as well as cancer in the respiratory tract and other tissues. Exposure to asbestos can also increase the risk of lung cancer, studies show.
“These findings have been repeatedly peer-reviewed by medical scientists since the first Report on Carcinogens was issued three decades ago, and they have consistently reaffirmed that asbestos causes cancer,” said Belluck, a partner in the New York asbestos exposure law firm of Belluck & Fox, LLP.

Some of the people most susceptible to asbestos exposure include those who work or worked in construction, shipyards, building demolition and automotive brake repair. Second-hand exposure is also a problem for the families of workers who could bring asbestos fibers home on their clothes and bodies. Veterans are another category of concern because the U.S. military used asbestos heavily from the 1940s until the 1970s.

Symptoms of mesothelioma — also called “asbestos cancer” — include:

  •     A persistent cough, which could be dry or could include mucous or blood
  •     Shortness of breath
  •     Problems swallowing
  •     Fatigue
  •     Weight loss
  •     Profuse sweating
  •     Painful breathing or pain the chest or ribs.
Belluck urged people to seek medical treatment promptly if they believe they have signs of mesothelioma. He also stressed that symptoms usually do not appear for 10 to 50 years after exposure to asbestos.
